PRODUCT DESCRIPTION:
The major interferon produced by mitogenically or antigenically stimulated lymphocytes. It is structurally different from type I interferon and its major activity is immunoregulation. It has been implicated in the expression of class II histocompatibility antigens in cells that do not normally produce them, leading to autoimmune disease. Interferon gamma is produced mainly by T-cells and natural killer cells activated by antigens, mitogens, or alloantigens. It is produced by lymphocytes expressing the surface antigens CD4 and CD8. The synthesis of IFN-gamma is induced, among other things, by Interleukin-2, FGF-basic, and EGF. Recombinant Rat IFN-gamma produced in E.Coli is a single, non-glycosylated, polypeptide chain containing 135 amino acids and having a molecular mass of 15609 Dalton.

FORMULATION:
IFN-g was lyophilized from 0.5X PBS pH-7.2.

RECONSTITUTION:
It is recommended to reconstitute the lyophilized product with sterile water at a concentration of 0.1 mgs/ml, which can be further diluted into other aqueous solutions.

STABILITY:
Lyophilized product is very stable at -20 degrees. Reconstituted material should be aliquoted and frozen at -20. It is recommended to add a carrier protein (0.1% HSA or BSA) for long term storage.

BIOLOGICAL ACTIVITY:
Recombinant Rat Interferon gamma is fully biologically active when compared to standard. The specific activity as determined by the cytopathic affect inhibition assay with murine NIH 3T3 cells chalenged with EMC virus was < 0.1 ng/ml, corresponding to a specific activity of 1 x 107 Units/mg.

PURITY:
Greater than 95% as determined by: Analysis by RP-HPLC. Reducing and non-reducing SDS-PAGE.

PROTEIN CONTENT DETERMINED BY:
UV spectroscopy at 280 nm. Analysis by RP-HPLC calibrated against a known standard. Quantitation on SDS-PAGE against a known standard.
